Takeda signs US$600M AI drug discovery deal with Insilico

In a strategic partnership, Takeda has joined forces with Insilico Medicine, based in Hong Kong, to leverage artificial intelligence (AI) in the initial stages of drug discovery within Takeda’s various therapeutic areas.

While the specific therapeutic areas and disease targets covered by this collaboration remain undisclosed, Takeda will gain access to Insilico’s Pharma.AI platform. This platform facilitates biological target identification, molecular design, and the prediction of clinical trial outcomes.

The primary focus of the collaboration will be the identification of potential drug candidates that align with predetermined scientific and early development criteria. Insilico will spearhead the AI-driven discovery efforts, while Takeda will be responsible for advancing selected candidates through clinical development.

Deal value and development rights

Under the agreement, Takeda will hold exclusive global rights for the development, production, and commercialization of innovative therapeutics discovered through this collaboration.

Insilico has disclosed that the deal encompasses approximately US$60 million in project initiation fees, immediate payments, and milestones. The overall value could potentially reach around US$600 million upon the achievement of preclinical, clinical, commercial, and sales-related milestones.

Additional payments are linked to various preclinical, clinical, commercial, and sales milestones, with Insilico also eligible to receive tiered royalties on future sales.

Alex Zhavoronkov, the founder and CEO of Insilico, mentioned that the proceeds from this agreement will support early-stage research and development activities within the collaborative program. He also noted that the timelines for later-stage developments will rely on Takeda’s clinical development initiatives and the coordinated efforts of both companies.

AI drug discovery partnerships

The Pharma.AI suite comprises tools geared towards target discovery, molecule generation, and the prediction of clinical development outcomes. Notable components include PandaOmics for target discovery, Chemistry42 for de novo small-molecule generation, and InClinico for forecasting clinical trial transition probabilities.

Insilico has also progressed its own AI-generated drug candidate, Rentosertib, formerly known as ISM001-055 or INS018_055, into clinical trials. This small-molecule TNIK inhibitor designed for idiopathic pulmonary fibrosis underwent evaluation in a Phase 2a randomized clinical trial.

Chris Arendt, the chief scientific officer and research head at Takeda, emphasized that this collaboration merges Takeda’s disease biology expertise with Insilico’s AI-driven discovery capabilities. Takeda is also integrating automation, robotics, and generative AI into its discovery processes.

This partnership with Insilico follows a previous AI drug discovery deal inked by Takeda earlier in the year. In February, Takeda embarked on a multi-year collaboration with Iambic valued at over US$1.7 billion, focusing on utilizing AI for the development of small-molecule drugs targeting cancer and gastrointestinal ailments.

Iambic’s platform features NeuralPLexer, an AI model employed for predicting the binding interactions between drug molecules and proteins.

According to data cited by the South China Morning Post from China’s National Medical Products Administration, Chinese drugmakers executed 157 out-licensing agreements totaling US$135.7 billion in 2025.

In the Takeda–Insilico collaboration, Takeda secures exclusive global rights to the candidates identified through Insilico’s platform. Insilico has reported signing collaborative agreements with a cumulative potential value exceeding US$7 billion since the beginning of the year.

Recently, Insilico announced a collaboration with South Korea’s SK Biopharmaceuticals, focusing on neuroimmune disorders. This agreement involves upfront and near-term milestone payments of up to US$18 million, with a total potential value exceeding US$2.5 billion.

In March, Eli Lilly expanded its partnership with Insilico in an AI-driven drug discovery agreement valued at up to US$2.75 billion. This deal granted Lilly exclusive global rights for the development, production, and commercialization of specific oral treatments then in preclinical development.

Following the announcement of the Takeda agreement, Insilico’s Hong Kong-listed shares surged by 13.5%.
